چکیده

To study the mechanism of 1-integrin function in vivo, we have generated transgenic mouse expressing a dominant negative mutant of 1-integrin under the control of mouse mammary tumor virus (MMTV) promoter (MMTV1-cyto). Mammary glands from MMTV1-cyto transgenic females present significant growth defects during pregnancy and lactation and impaired differentiation of secretory epithelial cells at the onset of lactation. We report herein that perturbation of 1-integrin function in involuting mammary gland induced precocious dedifferentiation of the secretory epithelium, as shown by the premature decrease in -casein and whey acidic protein mRNA levels, accompanied by inactivation of STAT5, a transcription factor essential for mammary gland development and up-regulation of nuclear factorB, a negative regulator of STAT5 signaling. This is the first study demonstrating in vivo that cell–extracellular matrix interactions involving 1-integrins play an important role in the control of milk gene transcription and in the maintenance of the mammary epithelial cell differentiated state.

Estrogen-induced mammary fibroadenoma in a lamb: hormonal and immunohistochemical aspects

A fibroadenoma was diagnosed in the mammary gland of a lamb by immunohistochemical method andmeasurement of oestradiol hormone. The tumor was characterized by an encapsulated firm mass with acreamy-white cut surface. Histologically, it consisted of variably-sized sinus ducts, covered by a single ormultiple layers of proliferated epithelial cells, and embedded in a loose connective tissue. Immun...

Interaction of primary mammary bovine epithelial cells with biofilm-forming staphylococci associated with subclinical bovine mastitis

Background: Staphylococci are recognized worldwide as one of the most important etiological agents of bovine mastitis due to their virulence factors such as their ability to penetrate inside mammary epithelial cells and their ability to form biofilm. Aims: The objectives of this study were to establish a model of primary mammary epithelial cells origin...

Expression of VLA1, VLA2 and VLA3 Integrin Molecules in Uterine Endometrium of Infertile Women with Unexplained Aetiology in Ahwaz-Iran

Background: Recent attention has focused on the expression of integrin molecules within the endometrium, and their relation to infertility. Objective: The present prospective study was undertaken to determine whether the endometrium of women with unexplained infertility differs in the expression of very late activation antigens (VLA) from the endometrium of normal fertile women. Methods: Thirty...

Loss of Panx1 Impairs Mammary Gland Development at Lactation: Implications for Breast Tumorigenesis

Pannexin1 (Panx1) subunits oligomerize to form large-pore channels between the intracellular and extracellular milieu that have been shown to regulate proliferation, differentiation and cell death mechanisms. These key cellular responses are ultimately necessary for normal tissue development and function but the role of Panx1 in development, differentiation and function in many tissues remains ...
